In vitro study | In cell culture, Nitazoxanide reduced parasite growth by more than 90%, with no signs of human and drug-related cytotoxicity. Nitazoxanide is a new parasite repellent of thiazolide, showing excellent in vitro activity against a variety of protozoa and helminths. Nitazoxanide and its metabolite tizoxanide are more potent than metronidazole against G. intestinalis, E. histolytica and T. vaginalis in vitro. Nitazoxanide showed potent inhibition of HBV and HCV replication. In replicon-containing cells of HCV, Nitazoxanide made subsequent treatment with Nitazoxanide plus interferon more effective, but did not enhance the effect of Nitazoxanide plus 2'CmeC. Nitazoxanide induces a decrease in the production of several HBV proteins (HBsAg and HBBeag, core antigens) in 2.2.15 cells, but does not affect the transcription of HBV RNA. Nitazoxanide showed a right to E. The IC50 and IC90 values of histolytica were 0.017 mg/mL and 0.776 mg/mL, respectively. intestinalis 0.004 mg/mL and 0.067 mg/mL, respectively, against T. vaginalis. 0.034 mg/mL and 2.046 mg/mL, respectively. Nitazoxanide was more toxic to Entamoeba histolytica than Metronidazole and Propimazole. |
In vivo study | In a sterile piglet model of diarrhea, oral treatment with Nitazoxanide(250 mg/kg) reduced the number of parasites after 11 days. Nitazoxanide induced a drug-associated diarrhea that may have affected its therapeutic efficacy. |
